Reverse Transcriptase PCR (RT-PCR) + Serology+ Viral Culture + Immunofluorescence
Reverse Transcriptase PCR (RT-PCR): Measles is caused by a single-stranded RNA virus, and the presence of its genome can be detected in clinical samples using RT-PCR. This test amplifies the viral RNA to detectable levels, making it one of the most sensitive and specific methods available.
Serology: Measuring IgM and IgG antibodies against the measles virus in the blood can help confirm recent or past infection. A positive IgM antibody test can indicate a recent infection, while a positive IgG test might indicate past exposure or vaccination.
Viral Culture: Although it's more time-consuming and less commonly done, isolating the measles virus from a clinical sample and growing it in cell culture can provide definitive proof of infection.
Immunofluorescence: This test uses antibodies tagged with fluorescent dyes to detect measles antigens in clinical samples.Direct or indirect immunofluorescence can be performed on nasal or throat swabs, or on blood samples.
Collecting samples during the acute phase of the disease, especially within the first few days of the rash appearance, can improve the accuracy of these tests.
