Blood Agar Overview
- Definition of Blood Agar
- Blood Agar is comprised of Tryptic Soy Agar supplemented with 5% Sheep blood.
- It provides a nutrient-rich environment for bacteria and is particularly useful for culturing fastidious organisms, which are microbes requiring strict physiological conditions.
- Blood Agar is also employed to differentiate bacteria based on their hemolytic characteristics, indicating the organisms' ability to lyse red blood cells (RBCs).
Hemolytic Characteristics
- Hemolysins
- Many types of bacteria produce secreted protein toxins known as hemolysins.
- Hemolysins are capable of lysing RBCs and destroying hemoglobin, which is responsible for the red color of blood.
- There are three major types of hemolysis:
- Beta (β) hemolysis
- Alpha (α) hemolysis
- Gamma (γ) hemolysis
Types of Hemolysis
Beta (β) Hemolysis
Description
- β-hemolysis results in the COMPLETE destruction of both RBCs and hemoglobin.
- It is characterized by a clear zone surrounding a bacterial colony on a blood plate.
Mechanism
- The toxin secreted by the bacteria diffuses through the agar, lysing all RBCs it contacts and destroying the hemoglobin within those RBCs.
- This absence of hemoglobin results in NO red color, leading to a visible zone of clearing around the colony.
Alpha (α) Hemolysis
Description
- α-hemolysis causes the PARTIAL destruction of RBCs and hemoglobin.
- This type of hemolysis is marked by a green coloration surrounding a bacterial colony on a blood plate.
Mechanism
- Certain bacteria secrete hydrogen peroxide, which diffuses through the agar.
- The hydrogen peroxide oxidizes the hemoglobin in RBCs to form methemoglobin, which has a green color.
- Thus, oxidized hemoglobin results in a green coloration, indicating α-hemolysis.
Gamma (γ) Hemolysis
Description
- γ-hemolysis signifies NO destruction of RBCs by the bacteria.
- It is illustrated as a non-hemolytic bacterial colony on a blood plate.
Mechanism
- There is NO effect on RBCs, hence NO color change occurs around the colony growing on the blood plate.
Summary of Hemolysis Types
- Beta (β) Hemolysis: Complete lysis of RBCs + hemoglobin; manifests as a clear zone.
- Alpha (α) Hemolysis: Partial lysis of RBCs + hemoglobin; results in green coloration.
