Parts of the Microscope

  • Base: The bottom support structure that stabilizes the microscope.

  • Arm: The part connecting the base to the head, used for carrying the microscope.

  • Nosepiece: Holds the objective lenses and allows for rotation to switch between them.

  • Objective Lenses: Different lenses with varying magnifications (4x, 10x, 40x, 100x) used to view the specimen at different levels of detail.

  • Stage: The flat platform where the slide is placed for observation.

  • Condenser: Focuses light onto the specimen, enhancing clarity and detail.

  • Ocular Lens (eyepiece): The remaginfication of a specimen formed by the objective lens

  • Body Tube Transmitter: Where the image is formed by the objective lens to the ocular lens

  • Illuminator: Light source

Magnification and Contrast

  • Magnification: The process of enlarging the appearance of an object, making it larger than its actual size. This is achieved through the combination of ocular and objective lenses.

  • Contrast: Refers to the difference in light intensity between the specimen and its background, which is essential for visibility. Adjusting the light source and diaphragm can enhance contrast.

  • Parfocal: A feature that allows the microscope to maintain focus when switching between objective lenses, minimizing the need for readjustment.

Objective Lenses and Their Characteristics

Types of Objective Lenses

  • Scanning Objective (4x): Shortest lens, used for initial scanning of the slide; color-coded with a red ring.

  • Low Power Objective (10x): Provides a wider field of view; color-coded with a yellow ring.

  • High Dry Objective (40x): Used for detailed viewing without immersion oil; color-coded with a blue ring.

  • Oil Immersion Objective (100x): Requires immersion oil for viewing; provides the highest magnification and resolution; color-coded with a white ring.

Color Coding of Objective Lenses

  • The color of the ring on each objective lens indicates its magnification level and type:

  • Red Ring: Scanning (4x)

    • Yellow Ring: Low Power (10x)

    • Blue Ring: High Dry (40x)

    • White Ring: Oil Immersion (100x)
