Types of Microscopes

  • Simple Microscope: Composed of one lens and provides relatively low magnifying power.

  • Compound Microscope: Composed of two or more lenses that provide powerful magnification.

Parts and Functions of a Compound Microscope

  • Illuminating Parts (Provide the light):

    • Mirror / Illuminator (Light Source): Reflects or provides light to view the specimen.

    • Substage Diaphragm / Condenser: Regulates and controls the amount of light reaching or striking the specimen.

  • Magnifying Parts (Enlarge the specimen):

    • Eyepiece (Ocular Lens): Magnifies the image (typically 10×10\times) and focuses light to the eye where the observer peeps through.

    • Objective Lenses: Gather light and magnify the image:

    • Low Power Objective (LPO): Magnifies 4×4\times or 10×10\times to locate and enlarge the specimen.

    • High Power Objective (HPO): Magnifies the specimen 40×40\times, 43×43\times, or 45×45\times.

    • Oil Immersion Objective (OIO): Magnifies the object 100×100\times.

  • Mechanical Parts (Support and connect components):

    • Body Tube: Holds the eyepiece and objective lenses.

    • Arm: Supports the tube, connects to the base, and is used for carrying.

    • Revolving Nosepiece: Holds and changes objective lenses.

    • Stage: Platform where the slide is placed.

    • Stage Clips: Hold the slide in place on the stage.

    • Coarse Adjustment Knob: Used for initial focusing.

    • Fine Adjustment Knob: Used for precise focusing.

    • Base: Supports the entire microscope.

Microscope Light Path and Magnification

  • Light Path Sequence: Light Source \rightarrow Condenser \rightarrow Specimen \rightarrow Objective \rightarrow Eyepiece \rightarrow Eye.

  • How It Works: Light from the source passes through the condenser and specimen. The objective lens magnifies the image, and the eyepiece lens magnifies it further to form a larger virtual image for the observer's eye.

  • Total Magnification Formula:   Total Magnification=Eyepiece Magnification×Objective Magnification\text{Total Magnification} = \text{Eyepiece Magnification} \times \text{Objective Magnification}

    • Example: 10× (eyepiece)×40× (objective)=400× total magnification10\times \text{ (eyepiece)} \times 40\times \text{ (objective)} = 400\times \text{ total magnification}
