Connective Tissue Overview

  • Definition: A diverse group of tissues that originate from mesenchyme of the embryo.

  • Functions: Provides cohesion to other structural elements and serves as the medium for blood vessel distribution.

  • Structure: Composed of widely scattered cells within an extracellular matrix (ECM) that consists of fibrous and amorphous components.

Components of Connective Tissue

1. Cells

  • Different connective tissues contain various cell types.

2. Fibers (Structural Proteins)

  • Include:

    • Collagen Fibers: Provide tensile strength.

    • Elastic Fibers: Allow stretching and recoil.

    • Reticular Fibers: Form a supportive net-like structure.

3. Extracellular Matrix (ECM)

  • Composed of ground substance, proteoglycans, glycoproteins, and water.

Cell Types in Connective Tissue

Fibroblasts

  • Characteristics: Most abundant; produces and maintains ECM, including collagen and fibers.

  • Staining Features: Nucleus is oval, dark-stained; becomes active in wound healing.

Chondrocytes

  • Functions: Maintains and secretes the cartilage matrix, found in lacunae.

Osteocytes

  • Roles: Maintain bone matrix, respond to stress, found in lacunae.

Adipocytes

  • Function: Store energy, provide insulation and cushioning.

Macrophages

  • Role: Engulf pathogens and debris (phagocytosis).

  • Originates from hematopoietic stem cells (HSCs).

Mast Cells

  • Function: Release histamine and heparin during allergic reactions; have many granules.

Plasma Cells

  • Function: Produce antibodies for immune defense; characterized by an eccentric nucleus.

Connective Tissue Fibers

Collagen Fibers

  • Appearance: Colorless, striated under the microscope; provides tensile strength.

  • Histological Staining: Appears pink in H&E; green in Masson's trichrome.

Elastic Fibers

  • Characteristics: Thinner and highly branched; allow for stretch and recoil.

  • Histological Staining: Stains pale pink in H&E; dark brown/black with orcein.

Reticular Fibers

  • Structure: Thin fibers composed of Type III collagen forming a branching network.

  • Function: Provide supportive frameworks for organs (lymph nodes, spleen).

  • Histological Staining: Stains black with silver stain; pink with PAS.

Ground Substance

  • Definition: A hydrated translucent gel where cells and fibers are embedded.

  • Key Components: Glycosaminoglycans (e.g. hyaluronic acid, chondroitin sulfate).

  • Function: Medium for nutrient and waste exchange; maintains tissue hydration.

Classification of Connective Tissue

1. Connective Tissue Proper

  • Includes loose and dense connective tissues.

2. Specialized Connective Tissue

  • Types: Cartilage, bone, blood, lymph, each with unique structures and functions.

Loose Connective Tissue

  • Characteristics: Loosely arranged fibers, abundant ground substance; provides support and cushioning.

  • Common Locations: Dermis, surrounding blood vessels, and within lymphoid organs.

Types of Loose Connective Tissue

  1. Areolar Tissue: Supports and cushions organs, aids in metabolic exchange.

  2. Adipose Tissue: Energy storage, insulation, and cushioning.

  3. Reticular Tissue: Forms supportive framework for immune cells.

Dense Connective Tissue

  • Characteristics: Highly fibrous, fewer cells than loose tissue, provides tensile strength.

  • Types:

    • Dense Regular: Parallel collagen fibers; found in tendons and ligaments.

    • Dense Irregular: Randomly oriented fibers; provides strength in multiple directions.

    • Elastic Connective Tissue: Contains high elastic fiber content; found in large arteries and lungs.

Specialized Connective Tissue

Cartilage

  • Semi-rigid, provides support with flexibility.

  • Types include: Hyaline, Elastic, and Fibrocartilage.

Bone

  • Rigid structure forming the skeletal framework; classified into compact and spongy types.

Blood

  • Fluid connective tissue responsible for transport and immunity; composed of plasma, RBCs, WBCs, and platelets.
