Tests for Starch, Reducing Sugars, Non-Reducing Sugars, Proteins, and Lipids in Food Samples

Starch Test for Food Samples

  • Purpose: To determine the presence of starch in a food sample.
  • Procedure:
    • Add a few drops of iodine solution to the sample.
  • Expected Results:
    • Positive Result: A blue-black color appears, indicating the presence of starch.
    • Negative Result: The solution remains orange-brown, indicating no starch is present.

Reducing Sugars Test (e.g. Glucose)

  • Purpose: To identify the presence of reducing sugars in a food sample.
  • Procedure:
    • Add Benedict’s solution to the food sample.
    • Heat the mixture in a water bath at about 80°C for 2–3 minutes.
  • Expected Results:
    • Positive Result: The color changes from blue (initial) to green, yellow, orange, or brick red, depending on the concentration of the reducing sugars present.
    • Negative Result: The solution remains blue, indicating no reducing sugars are detected.

Non-Reducing Sugars Test (e.g. Sucrose)

  • Purpose: To determine the presence of non-reducing sugars in a food sample.
  • Procedure:
    1. Boil the sample with dilute hydrochloric acid to hydrolyse the sugar.
    2. Neutralize the mixture with an alkali, such as sodium hydroxide.
    3. Add Benedict’s solution to the neutralized sample and heat again.
  • Expected Results:
    • Positive Result: A brick-red/orange precipitate forms, indicating the presence of non-reducing sugars such as sucrose.
    • Negative Result: The solution remains blue, indicating no non-reducing sugars are present.

Protein Test

  • Purpose: To test a food sample for protein content.
  • Procedure:
    • Add a few drops of Biuret reagent (which can be sodium hydroxide + copper(II) sulfate) to the food sample.
  • Expected Results:
    • Positive Result: A lilac/purple color forms, indicating the presence of protein in the sample.
    • Negative Result: The solution remains blue, indicating no protein is present.

Lipid Test

  • Purpose: To show the presence of lipids in a food sample.
  • Procedure:
    • Mix the food sample with ethanol, and then add water.
  • Expected Results:
    • Positive Result: A white, cloudy emulsion appears, indicating the presence of lipids.
    • Negative Result: The solution stays clear, indicating no lipids are present.
