Hypothalamus Overview
- The hypothalamus serves as a critical integrating center for various neural and endocrine influences that originate throughout the brain.
- It processes these influences and conveys messages to all parts of the body, functioning as a funnel that transmits the brain's effects on numerous homeostatic mechanisms to the body.

Anterior or Preoptic Group
- Preoptic nucleus

- Dorsomedial group

Connections of Hypothalamus
Afferents
- From limbic system:
- Involves the hippocampus and amygdala, connected to emotion and behavior.
- From cortex:
- Direct inputs from the frontal lobe, specifically area 6 and the prefrontal area.
- From olfactory lobe and paraolfactory area.
- Collaterals from the lemniscal system and fibers from the Reticular Activating System (RAS):
- RAS fibers are associated with alertness.
- From corpus striatum:
- It controls tone and involuntary movements strongly.
- From thalamus.
- From brainstem:
- Including the raphe nucleus, locus coeruleus, and medulla.
Efferents
- To brainstem and spinal cord:
- Efferents travel via pontine nuclei through reticulospinal fibers.
- Descending impulses terminate on lateral horn cells, regulating spinal sympathetic centers.
- The hypothalamus regulates blood pressure (BP), heart rate, and blood vessel diameter.
- To posterior pituitary:
- Fibers from the supraoptic and paraventricular nuclei form the hypothalamic hypophyseal pathway, ending in the posterior pituitary.
- To thalamus.
- To midbrain.
Functions of Hypothalamus
Regulation of Body Temperature
- The hypothalamus plays a vital role in maintaining body temperature.
Regulation of Anterior Pituitary Activity
- It influences anterior pituitary hormone release and activity.
Regulation of Posterior Pituitary Hormones
- Controls the secretion of hormones from the posterior pituitary.
Control of Circadian Rhythm
- It regulates daily rhythms in behavior and physiology.
Control of Sleep-Waking Cycle
- Plays a significant role in managing the cycles of sleep and wakefulness.
Control of Autonomic Nervous System (ANS)
- The hypothalamus is the head ganglion of the ANS, with stimulation leading to autonomic responses.
a) Anterior hypothalamus stimulation results in parasympathetic responses.
b) Lateral hypothalamus stimulation produces sympathetic responses.
c) Dorsomedial and posterior nucleus stimulation leads to increased adrenal medullary secretion.
Control of Hunger and Feeding
- Ventromedial Nuclei (VMN): Acts as a satiety center; lesions here can result in hyperphagia (hypothalamic obesity).
- Lateral Nucleus (LN): Functions as a feeding center; lesions lead to anorexia, often resulting in starvation.

- Gut peptides:

Role of Hypothalamic Peptides
- Increased Food Intake:
- Neuropeptide Y: Synthesized by neurons in the arcuate nucleus of the hypothalamus.
- Orexin-A and Orexin-B: Synthesized by lateral hypothalamic neurons.
- Ghrelin: Secreted by the stomach and hypothalamus.
- Decreased Food Intake:
- CART (Cocaine and Amphetamine Regulated Transcript).
- CRH (Corticotropin Releasing Hormone).
Regulation of Water Balance
- Regulation occurs in two primary ways:
a) Creating the sensation of thirst.
b) Controlling excretion of water into the urine.

- Stimuli:
- Hyperosmolarity and Hypovolemia.
- Mechanisms:
- Osmoreceptors in the hypothalamus stimulate thirst.
- Release of Antidiuretic Hormone (ADH) leads to increased water intake and decreased urinary volume.
- Results in restored osmolarity and blood volume.
- Additional factors:
- Angiotensin II and Baroreceptors contribute to volume regulation.
Control of Emotional Behavior
- Regulated by the limbic cortex.
- Two centers in the hypothalamus involved in emotional and behavioral regulation include:
- Reward Center: Located along the medial forebrain bundle, especially in the lateral and ventromedial nucleus.
- Punishment Center: Located in the medial hypothalamus and paraventricular zone.
Control of Sexual Behavior
- Sexual behavior is information processed by the anterior ventral hypothalamus.
