Citrate Test Notes
Citrate Test: Principle
- The citrate test assesses a microbe's ability to utilize citrate as its sole carbon and energy source.
- The test relies on the microbe's capacity to generate alkaline byproducts during citrate breakdown.
Citrate Test: Procedure
- Preparation of Citrate Agar Plate
- Use aseptic technique to prepare a Simmons citrate agar plate.
- Inoculation
- Apply the microbe being tested to the plate using aseptic technique.
- Incubation
- Incubate the plate at 37 °C for 48 hours.
- Observation
- Observe the plate for growth and any color change after incubation.
- Interpretation
- Positive Test: A color change from green to deep blue signifies alkaline products and indicates a positive result.
- Negative Test: No color change (remains green) indicates a negative result.
