The first method to test for gestational diabetes mellitus requires a two-step approach. In the first step, the patient completes a non-fasting 50-g glucose tolerance test. Plasma glucose is then measured after one hour. If this level is elevated, the patient is required to take a three-hour glucose tolerance test, which is the second step. This step requires a patient to consume 150 g worth of carbohydrates per day for three days while also fasting the night before their appointment. The patient then consumes the 100-g glucose tolerance test the following morning, and then the plasma glucose levels are measured at each hour for a total of three hours. ADA guidelines describe that two or more levels need to be elevated to diagnose gestational diabetes mellitus: fasting glucose is 95 mg/dL, one hour test is 180 mg/dL, two-hour test is 155 mg/dL, and three-hour test is 140 mg/dL

The second method used to test gestational diabetes mellitus requires a one-step approach. The patient is required to take 75-g glucose tolerance test after fasting the night before. The plasma glucose levels are monitored at each hour for a total of two hours. According to ADA guidelines, gestational diabetes mellitus is diagnosed if one of the following values are elevated: fasting glucose is 92 mg/dL, one-hour test result is 180 mg/dL, and two-hour test result is 153 mg/dL
